Ang-(1-7)可以抑制由Ang II上调引起的增殖细胞核抗原（PCNA）蛋白表达，A 779抑制Ang-(1-7)的该作用，但单独的A 779对诱导VSMC的增殖和迁移没有作用。 Ang-(1-7)预处理显著延缓Ang II诱导的VSMCs炎症反应，与上调的MCP-1，VCAM-1和IL-1β表达相关，Ang-(1-7)的这种作用也会受到A 779的抑制。但单独使用A 779对诱导VSMC的炎症反应没有影响。在VSMC中，用Ang-(1-7)预处理VSMC 5分钟显著抑制Ang II诱导的Akt和ERK1/2磷酸化，这种作用也被A 779阻断，但单独对诱导Akt和ERK1/2磷酸化没有作用[1]。 |

| 体内研究 (In Vivo) |
单独或联合输注Ang-(1-7)和A 779（400 ng/kg/min，s.c.）6周不能防止OVX大鼠的子宫萎缩或抑制体重增加。 A 779显著升高血清中骨特异性碱性磷酸酶（BALP），I型胶原蛋白（CTX），酒石酸盐抗性酸性磷酸酶（TRAcP 5b），骨钙蛋白（OC）和尿脱氧吡啶啉（DPD）的水平。输注Ang-(1-7)和/或A 779不会显著改变假手术组或OVX组中的血清矿物质浓度。相比于假手术组，OVX组中的AngII（P <0.05） ，AT1R（P <0.05），ACE（P <0.01）和RANKL（P <0.01）表达显著升高，而Ang-(1-7)，AT2R，ACE-2，MasR和OPG显着降低（P <0.01），当对OVX动物中给药A 779以后，发现AngII，Ang-(1-7)，AT1R，AT2R，ACE，ACE-2，Mas受体，RANKL和OPG蛋白表达没有变化。 阻断G蛋白偶联受体（Mas）的A 779可以显著抵消Ang-(1-7)对骨骼健康的有利影响，表明Mas受体在介导Ang-(1-7)骨保护作用中具有重要作用[2] 。 A 779（400 ng/kg/min）对Ang-(1-7)级联抑制效应可以显著消除captopril对骨代谢、矿化和微结构的保护作用。 A 779还可恢复OVX对RANKL表达和ACE-1/AngII/AT1R级联的影响，并下调OPG表达和ACE-2/Ang1-7/Mas途径[3]。 |
